Background:

 

The nucleosome is the basic subunit of chromatin and consists of 146 base pairs of DNA wrapped around an octamer of core histones (H2A, H2B, H3 and H4).

The 601 sequence, identified by Lowary and Widom, has high affinity for histone octamers and is useful for nucleosome assembly.

 

 

 

Application Notes:

 

Suitable for in vitro chromatin assembly and enzyme activity assays.

 

 

Details:

 

Recombinant nucleosomes were assembled by salt dialysis on 146 bp DNA from recombinant human core histones produced in E. Coli, purified using glycerol gradient and formulated in a storage buffer containing 20mM Tris-HCl pH 7.9, 1mM EDTA, 0.5mM PMSF, 1mM DTT and 40% glycerol.

Recombinant human nucleosomes are evaluated by native gel shift assay.
