Background:

 

The histone Variant H3.3 replaces conventional histone H3.1 in a wide range of nucleosomes in active genes and is incorporated into chromatin independently of DNA synthesis (1). H3.3 core Histones are variant nucleosomal core components were H3.1 is replaced by H3.3 (2). The H3.3 variant nucleosome consists of 147 base pairs of DNA wrapped around an octamer of core histone proteins H2A, H2B, H3.3 and H4 (3).

 

 

Protein details:

 

Recombinant human H3.3 core histones were produced in E. Coli, purified using FPLC and formulated in a storage buffer containing 20mM Tris-Cl pH 7.9, 1mM EDTA, 2M NaCl, 0.5mM PMSF and 1mM DTT. Protein concentration was determined by spectrometry.

 

 

Quality control:

 

Each lot has been evaluated by ESI-TOF analysis and 14% SDS-PAGE (NOVEX Tris-Glycine SDS, Invitrogen).
